Does anybody have an experience with stabilizing plant leaf tissues with RNAlater™-ICE Frozen Tissue Transition Solution? Is it necessary to homogenize the plant tissue for RNA isolation before using this solution?
It have been mentioned in the USER GUIDE (attached file) that: RNAlater®-ICE is a novel reagent for transitioning frozen tissue to a state that can be readily processed by common homogenization methodologies to extract high quality RNA. It circumvents the need to grind frozen tissue into a powder before homogenization, and even makes it possible to further dissect tissues before homogenization without RNA loss or degradation. We have used the normal RNAlater for collection and transportation of plant material (mostly leaves) from fields. We simply cut the collected leaves into small pieces and immerse them in the solution. Everytime we have got good quality RNA. Homogenization before preservation is not necessary.
RNA Later is used for the preservation of RNA in the tissue be it Animal or Plant. It does not effect the extraction protocol and the normal protocols are used to extract good quality RNA. Homogenization using liquid nitrogen is recommended though.
