Bullet kits which have insulin, FGF, FCS, normally used for fibroblast cell lines.

In my experience with dermal fibroblasts,the cells do well upto passage 10 in DMEM+10%FCS.After P10 they start to look more elongated and stressed.
You could probably use 2ng/mL of bFGF/FGF-2 or PDGF for the cells to do better.

Fetal fibroblasts grow pretty well for a number of passages without any GFs. Adult fibroblasts is different. Upto 4 ng/ml bFGF can help proliferation. In my experience, adding bFGF almost doubles the life span of the adult fibroblasts.
Thank you for all suggestions. I will try DMEM + 10%FCS + bFGF for adult fibroblasts. Because bullet kits are so expensive. Also, I will work with THLE-2 cell line. Do you have any suggestions for this cell line?
We also use DMEM with 10% FBS and penicillin 0.1 mg/ml, and streptomycin 100 U/ml without growth factors.
they may do better at lower oxygen with serum. 2-5% O2 works well and they do not get stressed out so quickly. Also we have found defined calf serum (a lot less expensive compared to fetal serum, about 1/4th the price). If you use up to 15% calf serum it works very well and is often better than fetal bovine serum.

Have you tried DMEM with 10% FBS and penicillin 0.1 mg/ml, and streptomycin 100 U/ml for THLE-2? Also I found LHC-9 medium (Gibco), it is suitable for bronchial epithelial cell lines but BEGM Bullet Kit is also suitable for them, may be LHC-9 can work for THLE-2. Do you have any idea about LHC-9?
