Does the hematopoietic stem cell population show any xenobiotic response to TCDD chemical? The hematopoietic precursor cell line had no impact of TCDD induction. Does this mean they have no response elements?

By TCDD response, are you measuring a reporter or target AhR target genes? Have you considered using phenol red free, reduced serum media for several days prior to TCDD induction? many nuclear receptors respond to chemical within the media, and that could give you lots of background. A colleague of mine uses glass containers so that bisphenol A from the plastics does not contaminate his ER reporter system.

Have you also looked at protein levels as well as mRNA levels? Levels of protein expression do not always correspond with mRNA levels. And therefore there could be a response in the cell lines that you are not detecting via using qpcr.
