I was thinking whether using NaBu might enhance my transfection effciency during a lentiviral production in 293Ts despite the fact my thrid generation system uses plasmids with no SV40 promoter.
As far as I know, Nabu is normally utilized when the plasmids used during transfection are under the SV40 promoter, but I've seen a number of papers using sodium butyrate while having plasmids with different promoters (RSV, CMV, etc) as well as arguing that this compound has beneficial effects other than enhancing SV40 transcription, such as increasing the number of cells able to express foreign DNA.
It seems clear that butyrate treatment of cells results in histone hyperacetylation, hence augmenting DNA availability and gene expression. Thus, will I achieve better transfection results using NaBu despite non-SV40 plasmids are used?
