In my experiment, I want to use serum (mouse) to test the creatinine level via HPLC.
1. During the preparation, I centrifuged the whole blood, and then transfer the supernatant (which is serum) to new clean tube.
2. Use 50% methanol to dissolve creatinine standard (1mg/ml).
3. Use HPLC to test creatine standard and sample (serum) .
I tried twice these days, but no result.
Therefore, I want to ask if someone use HPLC to test kidney function? Or some protocol recommended?

From your question I have understood that you have an interest in
creatinine blood test?!
If so, then HPLC does not appear method of choice.
For quantitative purposes you should use ESI-MS, because of it provides direct information in solution - blood in your case, furthermore at fmol to atomol concentration levels.
The quantitative procedure - our development based on stochatic dynamics with corresponding innovative formulas which we offer - yields to reliability 'r' higher than 0.99996 tested on mixtures of steroids. There are few examples of drugs, as well. The concentration levels of our tests are at pg.(mL)-1.
