Dear all.. I hope someone could enlighten me about my problem.
I've used Roche-Cell Death Detection Kit (Cat no:11544675001). I treated my TNBC cells with 2 different agents. At the beginning, calculations didn't have any problem and I could find the apoptosis fold change compared to the control group (solvent was DMSO). I did graphs in according to the enrichment factor and accepted DMSO as 1.
After this experiment, I used the same cell lines but changed one of the agent. But the solvent was the same (DMSO). I calculated the enrichment factor as same as the last experiment. But somehow I found my DMSO's enrichment factor was higher than the treatment group's enrichment factors. It was seem that DMSO was more toxic than my chemotherapeutic agents. In my opinion it is not normal. Am doing something wrong with the calculations? Is there anybody already used this kit and help me about the calculations please???
I'm stuck in this problem and couldn't find the reason and answer :(
