Are distorted cell cycle profile of cells treated at IC50 values for 24 hours due to high apoptosis (22% in one case and 43% in another case of test chemicals) acceptable? Can't I interpret the result as "at biologically active concentration (IC50), the test compounds exhibit cytotoxic effect rather than cytostatic mechanism?"

I guess that when preparing the cells for the cell cycle test, you used the entire contents of the microwells or flask without discarding the supernatant culture medium, which contains cell debris. As a result, the Sub-G1 percentage has been mistakenly reported as very high as the cell cycle arrest has not been detectable. I suggest first discarding the supernatant medium and then repeating the cell cycle test.
