Hello Eliane,

In my experience, you should perform DNA extraction prior to RAPD-PCR, to make DNA available to amplification.

A simple method can be used such as direct colony boiling in TE. 95°C for 15 minutes is usually enough, but depends on the type of bacteria you work with.
