I wish to differentiate monocytes from PBMC (patient's blood) into macrophage type M1 or M2.
I then want to evaluate their cytokines to make sure that they have transformed well into M1 or M2 type.
Now, by dint of doing literature, I find different information. So I do not know what to look exactly ...
Can you confirm or correct me according to what I remember?
After recovery of monocytes, I put them in RPMI medium, 1% FBS with M-CSF to transform them into macrophage. (I do not know how long ... 6 days?), Then I change the middle and put IFNg to turn them into M1 or IL4 to turn them into M2.
To confirm their differentiation, I look at the cytokines IL1B, IL6 or TNFa for M1 and IL10 for M2.
What do you think ? does that sound right?
