Besides ANSYS APDL and ANSYS ACP, if you use surface as your geometry, when you click "Model" in the "Outline" in ANSYS mechanical you see that two options will be available which are "Thickness" and "Layers" which you can use them for defining the stackup.

Using ACP just makes the process of defining stackup a bit easier, however, the result of all methods shall be the same.
