I am trying to put together dose response curves for both EtOH and Paracetamol using HepG2 cells and am unsure if they express CYP2E1. Some background seems to say they do not (MV5? cells). If not is there a liver cell line that does I can buy from ATCC?! Also, does anyone have any suggestions on what titrations to use that could match therapeutic doses ? 1mM, 10mM, 25 mM?
HepG2 as well as other liver derived cells line do have minimal or BQL CYP activities. More specifically CYP2E1 (measured as oxidation of Chlorozoxazone) may have very low activity and in some cases not measureable. I am not sure if your end-point is a toxicological target, bit if this is the case you should consider that using HepG2 you do have much smaller (compared to primary hepatocytes) metabolic drug clearance or metabolic activation (that is really important for the case of paracetamol).
The best in Vitro model are primary hepatocytes, of course are more expensive that HepG2 but now there are various commercial sources with price not prohibitive.
