For insect cells (SF9/High Five) we normally culture in SF900-III or Express Five serum free media, but I am looking at using cells cultured in Graces media for a specific experiment. I'm a bit confused though as to whether Graces media needs CO2 or not during culture. Gibco claim that an "ambient" CO2 atmosphere is essential to maintain pH, but does that essentially just mean that normal air is O.K and no additional CO2 is required. Every protocol I've read has said CO2 is unneeded.
