Has anyone ever used another medium than the "standard" RPMI + glutamine + FBS for the culture of EBV-transfected lymphoblastoid cell lines? RPMI would potentially interfere with our assay so we would like to use an alternative (other than PBS)...

An alternative (other than PBS) would depend upon your application. You can use Hank’s balanced salt solution (HBSS) and Earl’s balanced salt solution (EBSS) which are isotonic solutions used to maintain osmolality and pH in biological applications. Both HBSS and EBSS include glucose and sodium bicarbonate for short-term maintenance of cells outside of growth medium. The amount of sodium bicarbonate in EBSS is designed to maintain pH under 5% CO2, while HBSS has lower sodium bicarbonate levels and is intended for incubation without CO2. You can also use these two buffers with or without Ca2+/Mg2+.

EBSS would be appropriate. The incubation time will differ for different cell types. The duration ranges from 2-4 hrs. and can even extend up to 6 hrs. for certain cell types. I am not aware about the incubation time for LCL. But certainly not 24 hrs.Regards.
