Im agree with Lipnam, those are the first step in tissue and microbial culture , you can not prevent contamination withouth of using the good practice, but the Amphotricin B is a good anti yeast against most of Aspergillus and penicillum you should try it.

I think Lipnam suggestions are excellent one you should investigate a source of contamination either in the hood or the incubator. If i might add a point to his suggestion is to try to isolate your culture from other cultures in the incubator to prevent cross contamination from other cultures.

1. I'm not sure that fumigation can be happen or not? That is stem cell and animal clonong lab. when they got contaminat, they will not think it came from incubator... i will think come from person who work.

2. I already check media that i prepared ... still pure not contaminated.

3. I also use antibiotic/antimycotic (Pen/strp, Amphotericin B) at normal concentration, 3x and 5 x... still get contamination.

Anything that i can do... I'm so sad.

I would also say Lipnam's suggestion is the best. I work with stem cells and follow the same whenevr we have a contamination.. I would suggest you give it a try.
