We usually use TUNEL to detect apoptotic cell death before. But in macrophages, we found many tiny signals out of cells after LPS treatment. We consider that might be caused by necrosis or pyroptosis so the fragmented DNA flew out from broken membrane. (see figure attached). In addition, TUNEL positive also happens in necrosis and pyroptosis but we just want to watch apoptosis.

Could someone recommend a kit or reagent (we plan to use annexin-v conjugated with FITC and PI staining in situ) that is can be observed by fluorescent microscope without digestion for flow cytometry. (flow cytometry is not available for us).
