Hello, I'm phd student and on my work I seeded KM12/SM cell on 96 well (to detect the survival of those cell with a mutated gene) so I fixed those 96 plates with PFA 4% , stained with Cristal violet 0.1% and solubilized with acetic acid. I want to know if can I extract the DNA from those cell after all that process ?? . Thanks for everyone in advance.
Firstly I would like to thank you for your answer. I seed a high density of cell and I leaved them for 8 days of growth and there are now 75 000 cells and I treated them with ADC. The problem I don't have enoughth of drug that's why I didn't seed in other plate 1 million. And we wanted to explore the DNA that we have from the actual experiment
Even though you might be successful in isolating some DNA from such material, your data will not be considered meaningful as any findings can be attributed to fixation and not actual biological phenomena you are trying to study. You will need to purchase more ADC and perform the experiment again with living cells.
