If we raise the temperature of a histone-DNA solution, does the DNA unwind from the histones and eventually denatures? If so, could histones interfere in a PCR process?
During the DNA extraction process, 99% of protein was detected and the stage 1 of PCR is denature which was done at 95°C for from 30sec to 5 min. The protein were denatured all at this temperature except some high temperature proteins which is not histone.
