We always use HEK293A cells to package adenvirus, could HEK293T cells be used to do it?

HI there,
as far as I know, the HEK-A cells stably express an auxilliary plasmid that is needed to produce the adenovirus.
If you want to use HEK293T, you would have to co-transfect this plasmid into the HEK cells along with your target plasmids.
So short answer: I would stay with the HEK-A cells

Thank you very much, as I know, both 293A and 293T were coming from the original HEK293 cells, which stably expresses adenovirus related gene.
we use HEK293T cells as a packaging cell line for adenovirus in our lab.
we use the padeasy system developed by bert vogelstein lab.

You could use the cell line but you should have ectopic helper plasmid for the envelope. There are ready packaging cell lines, such as Phoenix derived from HEK293T cell line.

You can use 293T cells but as mention above it is recommend to add an ectopic palsmid for expressing the envelope proteins. For best result use a linear pEi (Polyethylenimine).
