I am having last few single use aliquotes of untransformed E.coli BL21(DE3) and E.coli BL21(DE3)pLysS competent cells from a used up cloning kit. Is there any specific growth medium or additional medium components instead of standard nutrient medium like Luria broth and of course chloramphenicol addition in case of pLysS competent cells?
LB is fine for growing overnight. If you want to freeze the cells after growing them then just add sterile glycerol to your grown cells for a final concentration of 15% then freeze at -80. The cell aliquot themsleves can be directly frozen at -80 (I assume they are already stored this way) since they either contain DMSO or glycerol.
Hanna Alalam is correct that you can just use whatever your standard growth media is, such as LB or YT etc. However I would suggest you streak out for single colonies and grow up a single colony to store away. I never liked growing populations with an unknown history.
