Does anyone know a sensitive and reliable apoptosis/cell death assay for Cos-7 cells? All the conventional plate based assays are sensitive for chemically induced cell death but when it comes to transfection induced cell death; a very sensitive one is needed.
I have tried Dioc6(3)/PI staining, TMRE /Draq7 staining, Annexin V-FITC/7AAD staining and the above mentioned individually followed by flow cytometry. All of the above give hardly any difference between live and dead cells when I can see morphologically the number dead cells is manifold higher than in control samples. I have tried to titrate the reagents up and down but nothing works on these cells. On Hela, MCF7, SkOV3, most of above work like a treat.
If anyone has tried any flow cytometry based assay(s) for COS7 or CV1 (untransformed COS7) cells, help would be very highly appreciated. If someone has a working protocol, that would be really wonderful. I shall be extremely grateful.
