I have a number of fungal cultures but have recently faced the problem to maintain them as they are all infested with mites. How can I recover my culture?
Recovering the cultures may be a bit difficult, but you can try re-suspending the spores in sterile Phosphate buffered saline and then filtering through a 0.1 mm filter. Re-inoculate the filtrate and incubate for some time. If there are still viable spores they may grow. For future preservation of cultures you may try overlaying with oil of Canarium schweinfurtii. We have tried this in our lab and results were good.
To recover your cultures without mites contamination I have used this way. Put the cultures inside a freezer for 24-48 hours. All mites and its eggs will die. After this, remove cultures from the freezer and grow your fungi normally, in culture media. That´s it!
you can look at your cultures unde a microscope, circle any areas that look clean and subb from there. if you have mites you need to look at how your cultures are stored...are these primary isolates? or these meant to be pure cultures, do you have any wood/ lead material n the same room where your cultures are?
I always trys and keep my cultures in double ziplocked bags inside a plastic container, you can throw a couople of moth/napthlalene balls in side the plastic container. dont keep plates for longer then a couple months...in saying all this despite your best intentions if you are working with fungi you will inevitably end up dealing with mitrs..good luck!
Fumigation of Laminar air flow chamber, incubators can be done to destroy mites.
Apart from this addition of Lindane powder (0.1g/L) to the medium will remove mites without affecting fungal growth.
Alternatively use polythene bags to store your cultures.
Use of napthalene balls is also recommended.
The culture infested with mites should be kept at -20 degree celsius overnight so that mites and their eggs loose their viability and next day subcultures can be done on fresh medium....Good luck.
I live in a tropical area from Mexico and we had a lot of problems with mites, we resolved using pills down the toillet its are cheap an a small piece in a box or tube culture is enough, mites and eggs can be destroyed, quickly, maybe in 15 minutes, the active ingredient is paradiclorobencene, its a little be toxic, but if you are carefully, it doesn´t matter anythig. Ok, I think the point was the traduction, it were refer to deodorants for toillet, you can cut a piece in small pieces with a hammer and putting inside a glas jar. About the toxicity is by contact or by eat , if you use a spatule or gloves is enough for your safety, I send you a link to see an imagen of the product.
You can maintain the fungal spores as suspension in sterile water, in closed flasks. Is a cheap and efficient method for periods of months, even years. Or store them in a freezer (minus 80 degrees Celsius), in 10% glycerol.
