Thanks Prof. for your response, Actually I am looking for the dilution of triton for such experiments. I am using the same amount of 5 ul based on various published protocols however I couldn't find the the percentage or dilution for triton. Let me know if you can help me with this. Thanks

as You mentioned: the optimum concentration of Triton x 100 for such method, 5 ul of Triton X 100 in 500 ul of plasma sample, you may use the dilution  triton exactly as Detergent (soap),

TBST(50mM Tris pH 7.5, 150mM NaCl, 0.05% tween -20) for regular westerns

I use 0.05% - 0.1% Tween-20 in PBS (PBST) or TBS . it is one of the most used recipe and works for almost all western blots, Triton X100 is stronger than Tween 20 and need less amount, I use Triton X100 at 0.05%

Triton X-100 is soluble in all proportions at 25°C in water, benzene, toluene, xylene, trichloroethylene,ethylene glycol, ethyl ether, ethanol, isopropanol, and ethylene dichloride.1 At 10% (v/v) in water, it gives a clear to slightly hazy solution
