My GeneJet Midiprep Wash Solution I was lost due to contamination with acetone (long story). I cannot find the composition in order to make up my own, and all I find in Invitrogen's website is Wash Solution I for RNA preps. I'm purifying plasmids; does anyone know if that solution is the same, or have ideas for replacement? Can I just use Wash Solution II and not worry about it?
The wash buffer appears to be along the lines of a final concentration of 5M guanidine hydrochloride and 25% isopropanol
Ricardo Azpiroz You cant find exact composition of the wash buffer anywhere as it is a proprietary product. And No, wash buffer II is not a replacement of wash buffer I
Buffer I is composed of chaotropic salt in water solution which is used to remove the protein and other salts, while wash buffer is just alcohol & salt in water solution to remove excess amount of salts. So buffer II cant be replaced by buffer I.
I suggest you to go though the trouble-shooting guide of the product. Your problem appears to be on user side. I could suggest you to try 5M Guanidinium thiocyanate in deionised water solution for the washing.
