Is it logical to co-culture two different cancer cell line (say SKOV3 and OVCAR3) to imitate the heterogeneity of the cancer alongwith the conditioned media of another noncancerous cell (say Fibroblast or Adipocyte) to check the effect on these on the TME ?

To imitate tumor heterogeneity, I would suggest you co-culture cancer cells with a prominent bystander cell type say for instance, mesenchymal stem/stromal cells instead of using another cancer cell line. Cancer heterogeneity progression involves interaction with multiple bystander resident cell types.
By doing so, it is possible that the observed morphologic and growth behavioral changes in successive co-culture could result from cancer–bystander cell fusion, as certain fusion hybrids may survive and proliferate to form heterogeneous progenies, each carrying certain traits of the parental cells.

So, you could try cancer cell fusion with bystander cells which would give you a direct route to tumor cell heterogeneity.
