I am interested in using a chitinase to degrade chitin in cell wall fragments (AIF) isolated from Aspergillus fumigatus. In this insoluble structure, chitin is covalently bound to beta-1,3-glucan and galactomannan and I want to degrade chitin without affecting the binding capabilities of either of the other components.
I have tried both chitinases currently available from Sigma-Aldrich Co. without success. What are your experiences using chitinases to degrade insoluble chitin bound in a structure?
You should transform chitin to colloidal chitin first so that to be accessible to the enzyme, normally through acid (hcl or phosphoric), i think it wont affect the attached groups (because when we did at lab we looked if it changes a lot in its aspects like degree of acetylation but not). Then should also consider the type of chitinase whether its endo or exo. But basically you need to make it colloidal.
