Hi all,

Just wondering if anyone has experience changing the extension temperature during the library amplification step in the Nextera XT protocol?

We're looking to sequence amplicons that are >85% AT-rich and my main concern is that the 72C extension temperature will likely cause poor amplification of the region. My thoughts being if I use 68C (or even 65C) it may yield better results?
