I was wondering the following;
I add a chemotherapy agent to cell culture media to make a solution. I store the solution in a 4°C fridge. I use the solution to treat cell cultures, with the goal in mind to measure the cytotoxicity of the chemotherapy agent added to the culture media.
Can the effects of the chemotherapy agent be diminished over time, when the agent is put into solution with culture media?

Vicky Dumaine The treatment has to be done using the Lethal dose effect of the specific drug (you can find that in PubChem). Likewise LD50 is generally used for the toxicity studies. And also the cell concentration has to be calculated based on the treatment.
It is preferable to add fresh chemotherapy solution to your media to avoid any variability. Of course you can store the culture media in fridge at 4C, however, it is not recommended to mix any chemical drug with your media if you are up to measure the cytotoxicity. As per @Rocio Campos-Vega, chemical properties of some agents might change due to different sensitivity. With lack of such reference, you might dislike to go ahead with your experiment and conducting a comparison study will be useful.
