I am trying to inoculate a fungal species into plant, so i am searching about the best protocol that help me to prepare fungal spore suspension and to treat the plant seedling with this suspension.
Thanks dear prof.Dr. Hani, I am trying to inoculate some fungal endophytes into maize plant in its seedling stage using root dipping method prior transferring the germinated seedlings to pots. I want to know:
1. What is the best solvent to harvest and suspend the fungal spores from a fungal culture ( Fusarium and Penicillium) plated on PDA medium?
2. What is the best way (Haemocytometer or spectrophotometer) to count and adjust the concentration of spore suspension?
3. What is the best concentration of spores that will be effective for maize inoculation?

1) Water should be fine. 2) Haemocytometers should be fine, although penicillium spores and the microspores of Fusarium will move under a coverslip better and provide more accurate spore counts than the macrospores of Fusarium, which are awkward. 3) The best concentration of spores depends on whether you are trying to mimic natural levels, or you want to make sure the plant gets infected. For endophytes, somehting like 1 x 10 6 might be right.
