We use E3019-3.5KU. We store at -20C, but after 3-4 times of opening, the esterase in the bottle loses its activity. We aim to use it in PBS buffer 7.4. Have anyone stored esterase in PBS buffer for a long time, of course at -20C? Please advise. Thank you.

First make single use aliquots to avoid freeze thaw cycles. Also consider storing at -80 degrees as the colder you store it at the longer it will last.

You can also store it in a 50% glycerol in PBS buffer. Since glycerol does not freeze at -20C, you'll be able to prevent degradation from freeze and thaw cycles. The safest option is always to aliquot your stock regardless of the storage buffer.
