I have decided to use a specific cancer cell lines for my project. However, mostly I found only B16- OVA expressing cell lines and that too gifted from a lab. How can I engineer OVA antigen into my cell line?
Not any cancer line. You'll want to make sure that you put Ova into a mouse line with the H-2Kb allele of MHC-I and/or I-A(d) of MHC-II. You won't get any presentation of your model epitopes if your tumor line of interest doesn't have them. A good guideline is to make sure that the line comes from the C57BL/6 strain, but others could potentially work too.
Also, remember, Ova is not a mouse protein. It comes from chicken. So there is no endogenous mouse locus that you can edit. If you choose CRISPR methods to insert Ova, you'll need to select an appropriate safe harbor locus to edit. You could also easily incorporate the gene using other methods such as retroviral vectors or drug selection of stably integrated plasmid transfectants.
