Which one is better for PCR optimization? If I work with GC-rich, do I need to use either of them?

You can use either of them. Both are reported to increase PCR efficiency and specificity. More people use DMSO as PCR additive. PCR buffers included in some commercial PCR kits are already optimized for PCR GC-rich template. So, it also depends on which kit you use. If the buffer in a commercial kit you use already optimized for GC-rich template amplification, then you don't need to add extra PCR additive. I use DMSO.
