Yes, we have done it before in my lab with HL60 cells in suspension. We followed a similar protocol to yours, but we used a plate centrifuge, so you can seed the cells already in the 96 well plates.
In theory, you could. You would have to spin the cell down and then add the MTT reagent. I am not sure how successful this would be though. I would do an alternate cell death assay that measures cell death in cell supernatants like G6PD release or ATP release. This way you could spin your cells down and use the harvested sups for the assay.
