For virus elimination, we use meristem culture, thermotherapy and cryotherapy in addition to other techniques such as using antiviral compounds. I am sure many of us have several explants in a plate of tissue culture media. So, if one of the explants is clean after the treatment/excision of meristem and others are still infected, will my clean explant get the virus by transmission through media? Which groups of viruses will reinfect this way and which ones are safe to work this way?

Theoretically it's possible. Try to give more investigation in pre-liminary stages etc. use 100% proved healthy initial material for in vitro establishment.

We have some clones that are infected with virus and needing virus eradication. Then we use in vitro-based techniques to remove virus. So my question is whether virus can reinfect a clean plantlet in tissue culture if it was cultured alongside an infected plantlet.

I would say that this would be an extremely rare event that may happen only with highly infectious viruses, like tobamoviruses.

In tissue culture sources of elimination are virus, bacteria and fungi that they can find in medium by water, explant and environment. Virus can't spread all of medium by one explant this kind of elimination is another source such as environment and water
Yes the virus can spread in tissue culture by contact between the plants if viruses mechanically transmission
The chances of spreading are very very low when the plants are not touching each other either by root or other means. I am just curious to know what kind of virus it is.

It depends on the type of virus. Potato viruses such as PVX are easily transmitted by contact between plants. So in case of PVX, it is possible if the plants are close enough to come in contact. Viruses such as PLRV, however, require a vector to be transmitted.

Thanks for your answers. We work with all kinds of viruses in our horticultural crops and our current work is with CMV and INSV in gentian, RBDV in Rubus and leafroll in grapes. oping to work on several viruses in hops as well.
