Just curious if there can be somaclonal variation in direct organogenesis or not; because in direct organogenesis i.e. from explant to shoot and then root occurs directly without the intermediate callus phase. But still everything occurs in artificial growth mediums; which I feel may have a tendency to induce somaclonal variation. Please explain and justify. Thank you
See the attached paper Figure 3. Somaclonal variation in Habanero Pepper (Capsicum chinense Jacq.) was observed through examine with ISSR Molecular Markers. Different band patterns were observed between the mother plant and the plants derived from direct organogenesis. So, somaclonal variation can occur for the plants derived from direct organogenesis.

SV can occur in all plant cells. A problem arises when SV is happening in a single totipotent cell that regenerates into a full plant resulting in a variant plant. If SV happens in a meristematic cell that is part of a bigger tissue, there is chance that this abnormal cell will be "selected away" by neighboring cells. (If not you will have a chimera) .
This is why SV is more dangerous with somatic embryogenesis, organogenesis, adventitious shoot formation,.... compared to axillary shoot formation where shoot are arising from organized tissues .
