If you are asking whether you could amplify the coding sequence of a gene from bacterial genomic DNA instead of plasmid with the same primers, you can, because bacterial genes do not have introns. Make sure that the sequence in the plasmid (for which the primers are made) is the same, does not contain any tags or so and that primers anneal on the cloned sequence and not on the plasmid itself.
