I have been trying to detect this protein since long time now. It's a 12 transmembrane containing protein that is of 70 kDa and also glycosylated. It localises in the plasma membrane of cells. My samples are BL6 mice kidney and long bone lysate and MC3T3 cell lysate. I am using Santa cruz biotechnology antibodies for probing on PVDF membrane. I have two Primary antibodyone that is against C-terminus of the protein and other that is against the TM domain of the protein. Lysis buffer that i have used is Triton-X 100/ RIPA. Sample preparation i have tried both at 95'C for 5 min and 37'C for 20 min. I load 100 or 50 ug of total cell lysate after quantification with bradford. Running buffer and transfer buffer is standard  1x tris -glycine buffer with 0.1% SDS and 1x tris-glycine buffer with 0.04% SDS and 20% methanol respectively. Blocking buffer used is 5% Skimmed milk (1hr, RT), primary (O/N, 4'C)  and secondary (1hr, RT) dilution prepared in 1x PBST (0.05% tween).

I get a lot of background and non-specific bands but not my band of interest.
