I am testing cytotoxicity of the molecules on the U251 and U87 glioblastoma cell lines. We did tests of the cytotoxicity with MTT and blue alamar, but since the molecules create radicals that affect mitochondria I was suggested to do cytotox that would is not based on the metabolic activity. Can you suggest the best based on your experience?
You can begin your analysis by simply counting your cells with tripan blue to assess viability.

I agree with Benjamin, simply doing the tripan blue exclusion assay would work. What you can also try, although in u87 cell this will be challenging, is the good old gold standard for toxicity testing in cancer cells - namely the colony formation assay.
