Can someone please share protocol for Apoptosis induction in MSCs (mesenchymal stem cells) by UV irradiation?? induction in other cell types can also be helpful. I am going to perform a short project on MSC apoptosis, if there are other methods other than UV which can help me please share them too. I’m really looking to find out what you’d consider to be the most efficient method as cost and time are limiting factors. thank u in advance!
We did exposure to UV in HaCaT and primary fibroblasts (hFB).
Exposure to UV-B(+UV-A) from trans illumination system (6 x 8W tubes UV-B) totally stopped cell doubling (without killing them) after more than 30 sec of exposure. 50 % of cytostatic effect was attributed to ~10 sec exposure (determined as metabolic activity by resazurin). See our publication.
No induction of apoptosis was found in HaCaT since this line is defect of p53 (we are not tested apoptosis in hFB). HaCaT cells died by mitotic catastrophe without uniform induction of apoptosis (no synchronisation and starvation in our protocol).
I believe you will find your appropriate exposure time (dose of UV) in MSC.
So you can use HaCaT as a control without correct apoptosis.

I cannot comment on the apoptosis induction by UV in MSC, but even with the ionizing gamma radiation we were not able to induce significant apoptosis in MSC isolated from bone marrov, dental pulp nor ligaments. Instead, these cells undergo pemature senescence.
