Drug preparation should be prepared in 1mL as a stock solution.Before treating with MTT it should be filtered through Syringe filter. Make sure you use wide range of concentrations of the drug to ensure the effect of drug over the cell growth, depending on the toxicity of the drug.

Hello, I think you should use 'amount in micrograms' instead of concentration. I agree with Mr. Rajendran, you should make a stock solution then dillute it in series of concentration (i used 5 series of dilutions)

If I understood your questions correctly, what you need is a drug solution of 10 mg/ml i.e. 10 ug/ul and add 10 ul of it to your well to get a final amount of 100 ug. I agree with the both the above answers that you need a stock eg. 10X = 100 mg/ml and then serially dilute it, say 2 times and add 10 ul of each dilution. By doing so your chances of getting exactly 100 ug drug in the well are more and you can assess the drugs potency better.
