I want to determine resistance gene expression when this particular compound am working with is applied on tomato seedlings and inoculated with a pathogen. Therefore need to collect some leaf samples to analyze. Am not quite sure of which leaves/leaflet I need to collect for this gene expression & biochemical analysis. This is my first time attempting this.
Please help
Flush and mature leaflets are to be sampled and measuring of the leaflet itself should be recorded in terms of providing wider surface areas

it depends on your project, the chemical you are applying, chemical mode of action, your pathogen and its mode action,
in addition, if the pathogen attacks particular region in plant,
generally speaking, always its better to take young fresh leaves for expression-related studies and in case if there are any symptoms, then those leaves which shows symptoms, but care should be taken not to collect very old, damaged or stressed leaves because then overall transcript level will be different and it will be very difficult to locate your desired transcript picture (or the one under question).
