Hi Reza, for my experince, if you just need the sample for TEM, you can just pick a sigle plaque into 500ul SM buffer(it will give you a concentration about 106 pfu/ml), then you can use it for TEM. At least it's ok for my phages. However, if the phage or the host have some products which have negtive effect on TEM, then you may need to clean your sample.
Hi Reza, for cleaner samples and better staining the phage should be pelleted and washed in 0.1 M ammonium acetate after PEG precipitation has been completed. This can be achieved using a fixed-angle rotor (e.g. 70Ti or F1010 Beckman rotors) at 25,000 g for 1 hour. If you're looking for really pure samples perform CsCl gradient centrifugation. Look for publications by H-W Ackermann for detailed methods.
