I'm running a soft agar experiment using an epithelial mammary cell line (MCF10A ER-Src). The bottom layer ( 2mL of 0.75% agarose/well) is prepared the day before while the upper layer is prepared at the time of the seeding of 2x10^4 cells/well using 1.5 mL of 0.36% agarose/well. The tubes containing the upper layer+cells are left with the remaining volume (I prepare it counting with one extra sample every time) were already solidified. Yet, the agar that is in the plates is shaking a lot and it seems not polymerized.
Can this be fixed still before the end of the 3 weeks (17th of June)?

If the agar is already dried out then your cells are likely dead.... shaking the agar will not prevent polymerisation, it will just decrease the size of the polymers but retain the viscosity.
You might want to put your plates in a box with a damp paper towel (change it every day to prevent mold). Or just upgrade your incubator to one with better humidity control.
