I am trying to express a single domain soluble protein with an N-terminal SUMO tag in E coli. As an alternative, I would like to express this fusion protein in mammalian cells. This will involve sub-cloning the coding sequence including the tag into a vector such as pcDNA. Is the SUMO-fusion polypeptide stable upon expression in the cytosol of mammalian cells? Or is there a different sequence of SUMO that should be used for mammalian expression? Thank you.
