Please, I want to know if JEG-3 cell line can be a good model for assessing the effects of environmental chemicals on progesterone and estradiol production. I will also appreciate suggestions on other cell lines that can as well serve as in vitro models for this.
You need to research the cell line to see if it has an endogenous receptor for your target. If not then you are looking at transforming the cells with the receptor. I would look at this cell line and others that have the endogenous receptor.
Ukpai, I think that the choice of cell line to use really depends on the question that you want to address. In your posted question, it sounds like you may want to determine if a test chemical with unknown activity is able to affect progesterone and estrogen output. I think that it might be better to use two different culture methods. I would use a cell line that is known to express aromatase to assess the effects of your test chemical (plus/minus an anti-estrogen) on the ability of the cells to produce estradiol. To assess the effect of the test chemical on progesterone output, I would use luteinized granulosa cells from either mice or human granulosa cells obtained from an IVF clinic. Thanks John and Warren for your suggestions. I have been able to do some literature search and I can confirm that JEG-3, Bewo and H295R cell lines as well as primary placenta cells and ovarian granulosa cells can be used for the assay.
I have used the H295R cell line and primary placental cells with success. Granulosa cells are a little tricky. Since there is already an established literature using the H295R cell line for aromatase activity you may want to start there so that you don't need to re-invent the assay and only need to optimize for your own lab.
