MTT assay was performed on various cell lines and IC50 was calculated. However, cell viability was not performed separately. Is it logical to show by any means that compounds under study were not affecting cell viability if IC50 values are larger (less potent)?
I would suggest, that MTT assay is not the best assay to determine IC50 anyway. Why dont your try PI staining and measure apoptosis to get a clearer result. MTT assay does not really show proliferation, whereas it is often used in this way.
I'm agree with Christian Müller. compare MTT assay, I suggest to analyze using PI/annexinV and we get the % viable cell, early/late apoptosis, necrosis.
but sometimes, researcher use MTT assay to analyze the viability cells. may I know, do you put the + control or solvent control (medium only, solvent to dissolve your sample)? Do you see the formazan crystals?

If you only just want to determine IC50 values, MTT is a good tool. But if you want to study mechanisms of action, MTT does not work since it is an indirect way to quantify viability
