Guys, I have a High capacity cDNA reverse transcription kit, but it has been expired for 5 months. I would like to konw if the enzime is working yet. Should I buy a new kit?
Depends on the goal of your experiment. For feasibility studies or exploratory data, sure. But if you are trying to generate repeatable data for a figure or a journal, you may have some difficulties getting the same values. It may be just fine, if it hasnt gone through too many freeze-thaw cycles.
I'd say it also depends on how hard it was to get your starting materials. If your samples are very limited/expensive/precious to create, just buy a new kit.
I would propose to buy a new kit, since polymerase activity and efficacy strongly depends on age, thaw/freezing sequences/times etc.
However, using this kit should also result in cDNA (but to unknown extent//efficacy, which makes possible results not easy to interpret compared to other results using a fresh kit).
I am also facing the same problem regarding the cDNA kit @ Erika .
I am not getting the desired values in my real-time PCR experiment. Is this happening becoz of an expired cDNA kit ?????? please help me by answering a good solution. I am not getting good bands in my beta-actin RT PCR. so may be becoz of the expired kit.
