I make DNA extraction of vaginal swab brucella then Amplification using specfic primers for Brucella meltinesis but the result was no pcr products
no any band
when I search I was found I MUST culturing .
but I need an alternative method to extract DNA directly from swab without culturing?
I would not try and PCR off the swab itself as it will have other things which will inhibit the reaction itself.
You could try taking the swab and adding either a small amount of PBS or a transport medium (200ul - 1ml), squeeze the swab in the medium to get all the contents out into the medium and then perform a nucleic acid extraction on the solution. Then perform PCR on the resulting eluates.

without EXTRACTION is not safe because , it is sure it has an inhibitors?
