Well currently I am doing research on plant extraction. The plant powder has to be dissolved in different solvents such as Hexane, EA, mixture of EA-CH3OH and methanol at ratio 1:50. My issue is after extraction my crude product for hexane very less around 2 mg compared with other solvents. This crude product need to be sent for proton NMR analysis. I read the minimum weight of sample for H-NMR is 5 mg to get good spectra. My question is can I increase the weight of solute and solvent but still stick to 1:50 ratio? For example currently I use 0.1 g of solute and 5 ml of solvent. I want to increase to 0.3 g solute and 15 ml of hexane but other solvents still stick to 0.1 g solute and 5 ml of solvent. I have limited samples.
