I have a protein complex that binds to Ni-NTA Resin. If I elute with Imidazole, it needs about 150 mM of it. The 150 mM imidazole strips off some of the proteins from the complex as well, resulting in a heterogenouse complex. I am trying to find a way to elute my complex with a different, more gentle method, like EDTA. However, my complex also contains a Zn++, for which I intend to use 1 mM EDTA or less. Could anyone with experience using EDTA elution for His-Bi-NTA share your expertise?
