Before ask, My scheme is

'COOH particle-NH2 oligonucleotide conjugation'->'DNA:DNA hybridization'

In hybridization, use saline sodium citrate buffer because It provides ion(salt) for stronger bind DNA. But I do hybridization after amine DNA conjugation, so I use to SSC buffer in amine coupling.

Is it OK use SSC buffer for amine coupling? it NOT inhibits amine coupling?

ps. Can I use PBS buffer in hybridization? (glibco PBS)
